Genetic Factors in Schizophrenia Risk
نویسندگان
چکیده
Recent findings Multiple and variable genetic and environmental factors interact to influence the risk of schizophrenia. Both rare variants with large effect and common variants with small effect contribute to genetic risk of schizophrenia, with no indication for differential impact on its clinical features. Accumulating evidence supports a genetic architecture of schizophrenia with multiple scenarios, including additive polygenic, heterogeneity, and mixed polygenic-heterogeneity. The epigenetic mechanisms that mediate gene– environment (GxE) interactions provide a framework to incorporate environmental factors into models of schizophrenia risk. Environmental pathogens with small effect on risk have robust effects in the context of family history of schizophrenia. Hence, genetic risk for schizophrenia may be expressed in part as sensitivity to environmental factors.

Additive effect of MTHFR and GRIN1 genetic polymorphisms on the risk of schizophrenia
Schizophrenia is a complex disorder with polygenic inheritance. The MTHFR gene (OMIM: 607093) plays an important role in the folate metabolism. It has been suggested that C677T (rs1801133) and A1298C (rs1801131) genetic polymorphisms in the MTHFR gene lead to the decreased activity of the methylenetetrahydrofolate reductase enzyme which may have significant effect on developing schizophrenia. W...
متن کاملHyperprolactinemia and CYP2D6, DRD2 and HTR2C genes polymorphism in patients with schizophrenia
Introduction: Hyperprolactinemia is a common serious side effect of antipsychotic medications that are currently used in the treatment of patients with schizophrenia. Pharmacogenetic approaches offer the possibility of identifying patient-specific biomarkers for predicting the risk of this side effect. We investigated a possible relationship between variants (SNPs) in genes for cytochrome 2D6 (...
متن کاملSchizophrenia, Inflammation and Toxoplasma gondii Infection
Genetic analysis suggests that schizophrenia is a polygenetic disease, mostly consisting of common mutations with low risk and rare mutations with higher risks [1]. Although data suggests a strong genetic component to the risk of developing schizophrenia, environmental and/or developmental risk factors also contribute. Currently, there are no interventions that target and mitigate the effects o...
متن کاملPrenatal Factors in Schizophrenia
The purpose of this review is to summarize the current state of knowledge on how nongenetic factors occurring before, during, or soon after birth are related to schizophrenia. Schizophrenia is a complex psychiatric illness with a varied clinical presentation that has both environmental and genetic origins and that may result from insults to the nervous system that occur throughout development. ...
متن کاملGenetic principles and methods in high-risk studies of schizophrenia.
Recent advances in genetic epidemiology present excellent opportunities for future high-risk studies of schizophrenia. Improved methods are now available for specifying the natural boundaries of the schizophrenia spectrum and evaluating the mode of inheritance of schizophrenia and its biosocial risk factors. Path-analytic techniques permit the derivation of multifactor indices of both biologica...
